Van der Vaart sustained the injury in his left leg in the closing stages of last Sunday's 5-1 home defeat against Manchester City.
The Dutch international has since been diagnosed with a hamstring tear after a scan and is not expected to return until the middle of October.
That means Van der Vaart will miss Premier League games against Wolves, Liverpool, Wigan and the north London derby against Arsenal at White Hart Lane.
He will also be unavailable for the start of Spurs' Europa League campaign, including games against PAOK Salonika and Shamrock Rovers.
"An MRI scan has confirmed that Rafael van der Vaart has suffered a hamstring tear," a statement on Tottenham's website confirmed.
"The Holland international injured his left hamstring in last weekend's home match against Manchester City and is expected to be sidelined for up to six weeks."
