The friendly will mark the opening of their new 22,500-seater Amex Stadium, more than 14 years after the final game at their previous home, the Goldstone Ground.
Poyet said: "I've been looking forward to this day since the day I got the job because everyone was talking about this moment.
"I'm not surprised the tickets have sold out because I know how much this stadium means to the whole city.
"On top of that we are playing Spurs so it's going to be a very special day for everyone and I hope we give the fans a lot to cheer about."
