Anthony Gardner suffered a hairline fracture of his fibula in Sunday's FA Cup quarter-final at Chelsea and Ricardo Rocha is ineligible for the UEFA Cup.
Martin Jol said: "Anthony came on at Chelsea and after a couple of seconds took a knock to his knee. We'll do a CT scan but it looks like he'll be out for around six weeks.
"It's a blow but it's also a challenge because we have players who can play in one or two positions. Fortunately, we have a squad of versatile players who can cover our defensive positions. We also have Charlie Lee, captain of our reserves, in the squad as well."
"We have a versatile squad, Pascal Chimbonda and Didier Zokora can both play in the centre and, if necessary, we also have Tom Huddlestone.
Spurs will also be without goalkeeper Paul Robinson (thigh), Jermaine Jenas (groin), Ledley King (foot), Danny Murphy (achilles) and Benoit Assou-Ekotto (knee) against Braga this Wednesday.
